Pseudorabies virus displays variable numbers of a repeat unit adjacent to the 3' end of the glycoprotein gII gene.

چکیده
The coding region of the glycoprotein complex gII of pseudorabies virus (PRV) is located in the unique long part of the genome on SalI subfragments 1A and 1G of BamHI fragment 1 (map units 0.105 to 0.130). Fragment 1G which includes the 3' end of the gII gene displays a size heterogeneity among different PRV strains and also within plaque isolates of a given strain. To reveal the cause of this heterogeneity and whether it might affect the gII-coding region we sequenced different 1G fragments of the PRV strains Ka, Phylaxia and Dessau, and determined the 3' end of the gII mRNA by S1 analysis. These data show that the size heterogeneity is caused by the presence of a variable number of tandemly repeated DNA sequence downstream but adjacent to the coding region of the glycoprotein gII gene. The 3' end of the gII mRNA was mapped about 24 bp upstream of the first repeat unit. A 15 bp sequence 5' GGGACGGAGGGGAGA 3' is repeated from three to over 50 times in different 1G fragments. It is the only repeat unit present in strain Ka, whereas the Phylaxia and Dessau strains show additional modifications.

Background: Little knowledge of synonymous codon usage pattern of pseudorabies virus (PRV) genome, especially the UL31 gene in the process for its evolution is available. Objectives: In the present study, the codon usage bias between PRV UL31 sequence and the UL31-like sequences was identified. Materials and Methods: We used a comprehensive analysi...
متن کاملMolecular Genetic Analysis of the Variable Number of Tandem-Repeat Alleles at the Phenylalanine Hydroxylase Gene in Iranian Azeri Turkish Population
Background: The variable numbers of tandem-repeat (VNTR) alleles at the phenylalanine hydroxylase (PAH) gene have been used in carrier detection and prenatal diagnosis in phenylketonuria families. This study was carried out to analyze VNTR alleles at the PAH gene in Iranian Azeri Turkish population. Methods: In this study, 200 alleles from general population were studied by PCR. Results: The fr...
متن کاملPutative glycoprotein gene of varicella-zoster virus with variable copy numbers of a 42-base-pair repeat sequence has homology to herpes simplex virus glycoprotein C.
A strain variation of varicella-zoster virus that maps to the UL region of the genome was found to be due to different copy numbers of a high GC 42-base-pair repeat. DNA sequence analysis of this variable region showed the sequence to be 5-GCGGGATCGGGCTTTCGGG(A/T)AGCGGCCGAGGTGGGCGCGACG-3. Strains Scott and Webster both contain 7 and 32/42 copies of the repeat, whereas strain Oka has exactly 4 c...
متن کاملConstruction of an Eukaryotic Expression Vector Encoding Herpes Simplex Virus Type 2 Glycoprotein D and In Vitro Expression of the Desired Protein
To construct of an eukaryotic expression vector encoding herpes simplex virus type 2 (HSV-2) glycoprotein D (gD2), an Iranian isolate of HSV-2 was propagated in HeLa cell line and its DNA was extracted and used as template in polymerase chain reactions (PCR), to amplify gD2 gene. Primers were designed and the restriction enzyme sites for EcoRI and XhoI were considered at their 5′ ends respectiv...
متن کاملProtection of mice and swine from pseudorabies virus conferred by vaccinia virus-based recombinants.
Glycoproteins gp50, gII, and gIII of pseudorabies virus (PRV) were expressed either individually or in combination by vaccinia virus recombinants. In vitro analysis by immunoprecipitation and immunofluorescence demonstrated the expression of a gII protein of approximately 120 kDa that was proteolytically processed to the gIIb (67- to 74-kDa) and gIIc (58-kDa) mature protein species similar to t...
